BJ PL01 Keeping a lab notebook

Objective: Making an organized log of labs and keeping a good lab notebook. Objectives can be
as long or short as you would like. Reminds what we were doing during experiment.

Procedure:

1. Steps of any lab procedures. What kind of chemicals, temperatures, growth
conditions, etc.

2. Any deviations that may occur doing the experiment will be made red

3. Should be numbered steps

Notes:

Basic equipment

-Cleaning: autoclave 125 degrees, uses heat, pressure and steam to sterilize environment.
Steam is more efficient than hot air (steam sterilizes in minutes while air could take hours). Put
materials in to sterilize before and/or after lab.

-Growing: Fixed incubator, chamber to store samples, airtight (37 degrees C) and shaker
incubator, shakes to aerate samples to help bacteria grow (37 degrees C)

-Visualizing: Microscopy

-Storing: Refrigerator at 4 degrees C (slows bacteria from growing)

-Lab safety: never eat or drink in the lab

-PPE

Latex gloves, niatrial gloves (hypoallergenic), cold gloves (liquid nitrogen or cold), thermal
gloves for heat (pulling out of autoclave), lab coat

Never leave lab with any PPE on! Not even hallway or bathroom.

Course Objectives: cultivation of samples (growth conditions), identification of samples
(biochemical assays), evaluation of samples (microscopy)

BJ PL02 Introduction to Microscopy



Objective: To become familiar with the basic components of a light microscope as well as how
to load a sample for viewing.



Procedure:

1. Review parts and components

2. Load sample slide onto microscope

3. Select magnification

4. Make necessary adjustments to optimize sample visualization



Notes:

Lens makes object larger, an object to look at, and a light source. With these three things you
have a microscope.

Basic parts: eyepiece, arm/neck, objectives, revolving nosepiece, stage, stage controls, stage
clips, coarse/fine focus, iris diaphragm, light source, base.

Our microscope today is standard light microscope.

Oculars, you can adjust to look. If you see overlapping circles you need to adjust. Always
move microscope by putting a hand on the arm and under the scope. These also have
magnification (10x in our lab)

Lenses-provide magnification. 4 different objectives and you can grab and rotate to change.
Shortest objective has a 4X magnification, then 10X, then 40X, and finally the 100X (largest).
